in 60 minutes after the smear preparation . 4) Expose the smears to air for 1 hour to remove formalin. 5) Prepare the incubating solution as follows . Fifty mg of a-naphthylphosphate, 50 mg of Fast-Garnet-GBC salt, 50 ml of acetate buffer (pH 5.0, 0.1 M) are mixed, shaken well , and filtrated. 6) Incubate in it for 90 minutes at 37•Ž. 7) Wash well in distilled water . 8) Counterstain nuclei with Meyer's hematoxylin for 30 minutes. 9) Wash for 5 minutes in tap-water.

Histochemical demonstration of acid phosphatase activity in various blood cells.

Acid phosphatase activity of various blood cells were histochemically observed on peripheral blood and bone marrow smears using the azo dye method which had been reported in my previous paper, and following results were obtained. 1) In the neutrophilic granulocyte series, the largest acid phosphatase score was obtained in immature cells, which might include promyelocytes, myelocytes, and metamy...

A histochemical procedure for localizing and evaluating leukocyte alkaline phosphatase activity in smears of blood and marrow.

By LEONARD S. KAPLOW T HE RESULTS of previous studies’8 on the histochemical localization of alkaline phosphatase activity in cells of blood and bomie marrow have been varied and contradictory. All such studies, except that of Vercauteren,8 were based on the Gomori-Takamutsu procedure.” ‘#{176}Numerous investigators”9 have shown that this procedure may produce false staimiing of nuclei and othe...

Diazophthalocyanins as Reagents for Fine Structural Cytochemistry

This paper reports the synthesis of 14 diazophthalocyanins containing Mg, Cu, or Pb as the chelated metal. To assess the usefulness of these compounds for fine structural cytochemistry, the relative coupling rates with naphthols were tested as well as the solubility of the resulting azo dyes. Three of the diazotates were reacted with tissue proteins in aldehyde-fixed material, and the density i...

An ultrastructural study of acid phosphatase localization in Phaseolus vulgaris xylem by the use of an azo-dye method.

The localization of acid phosphatase during xylem development has been examined in the bean, Phaseolus vulgaris. The azo dye, the final reaction product, is initially prominent in the dictyosomes, vesicles apparently participating in secondary wall formation, and in the middle lamella of the young vessel element. Final reaction particles are also present in mitochondria, chloroplasts, and certa...

The colorimetric determination of phosphatases in human serum.

There are a number of methods currently available for the quantitative estimation of acid and alkaline phosphatase activity in biologic materials (l-5). In all the techniques, a phosphoric acid ester serves as a substrate and calorimetric determinations are made either of the inorganic phosphate or of the organic moiety of the substrate released by enzymatic hydrolysis. A description of a new m...
